Manuel Calderon is the younger brother of a former drug lord, Cesar Calderon, and murderer of Dr. Valerie Monroe.
Monroe was taking care of his brother Cesar following a recent heart attack. The two were gradually becoming romantically involved as well, to the point that Cesar gave Monroe a bracelet that had belonged to his mother (although he denied that their relationship was ever physical).
Manuel discovered that Dr. Monroe was in touch with state prosecutors. Believing (incorrectly) that she was an informant working to destroy his brother, Manuel murdered her. He bludgeoned Monroe into unconsciousness, then injected air into her carotid artery, causing an air embolism which killed her.
Manuel attempted to dump the body by placing it into the coffin of a man who was about to be buried, but the pall-bearers dropped the casket and Dr. Monroe's body was revealed. He kept his act secret from his brother because he believed that Cesar was blinded by his affection, but the subsequent investigation revealed that Monroe had actually been investigating drug theft at her hospital, and her contact with the prosecutors had nothing to do with Cesar.
